طريقة طيفيه بسيطة وسريعه وقابلة للتكرار لتقدير الفاموتيدين تعتمد الطريقه على تفاعل الننهيدرين مع الأمينات الأوليه الموجوده في الفاموتيدين حيث يحدث التفاعل في وسط قاعدي باضافه (NaOH) ينتج عن هذا التفاعل تكوين مركب لونه ارجواني عند الطول موجي الاعلى ( 550 نم) , حيث يتطلب التفاعل درجة حراره تصل الى حرارة 80 درجة مئوية ورقم هيدروجيني (10 ) ولمدة دقيقتين . المنحنى المعياري الناتج خطي مستقيم كما هو موضح بالمعادلة (1015.7x - 0.0152) مع معامل ارتباط مرتفع قدره (0.9921= 2R) . وتم الحصول على معامل امتصاصية بمقدار . (1015.7 L /mol /cm) كما تم حساب قيمة الحد الأدنى للكشف والتكوين للمركب الناتج فكانت LOD تساوي 0.00915 ?g mL و 0.031 ?g mL LOQ أيضا تم حساب مقدار نسبة التغير في الخطأ في التجارب من خلال إعادة العمل للتجارب في اليوم نفسه أكثر من مرة فكانت اقل من % 0.39 والدقة في نسبة الانحراف المعياري (RSD%) تتراوح ما بين % 0.4الى % 0.6 وإعادته خلال أيام متفرقة في خطأ نسبي اقل من % 0.38 والدقة في نسبة الانحراف المعياري (RSD%) تتراوح بين % 0.13الى .% 1.13

 A simple sensitive and reproducible method to determine Famotidine in injection sample based on the formation of a purple color product with ninhydrin was investigated in basic media using NaOH, the product was measured at the best ?max at 550 nm. The reaction occurs at temp 80?C and pH 10.0 and its equilibrium time was established in 16 minutes, the obtained calibration curve is linear over the range of concentration (2.04?10-4 - 5.11?10-4) mol/L with regression equation of A = 1015.7x - 0.0152 with correlation coefficient R? = 0.9921. The molar absorptivity value =1015.7 L.mol-1.cm-1. The limit of detection (LOD) and limit of quantification (LOQ) are calculated to be 0.00915 ?g mL-1 and 0.031 ?g mL-1. The intra-day accuracy expressed as relative error less than 0.39 % with precision (RSD %) ranging from 0.4% to 0.6%. The inter day accuracy ranged (RSD %) from 0.13% to 1.13% with a relative error less than 0.38 %. The stoichiometry of the reaction determined molar combining ratio 1:1 between Famotidine and ninhydrin. The initial rates of the experimental reactions with Fam are follow a pseudo-first order, plot for log k vs log [Fam]
